Utilisation des marqueurs microsatellites pour l'étude des contributions parentales chez l'huître creuse Crassostrea gigas ArchiMer

Numerous aquacultural species show very high fertilities. As a consequence, it is possible to produce, in hatcheries, a large amount of sibs from a small amount of parents. The sibs' genetic variability is, of course, directly linked to the number of parents involved, but it may be largely lowered by imbalanced parental contributions. Polymorphic genetic markers are very handy tools in the study of these phenomena in cultivated populations (Harris et al. 1991; Herbinger et al., 1996a; Garcia de Leon et al. 1998) or in natural populations (Ferguson et al, 1995; Kellog et al., 1995; Colbourne et al., 1996; Herbinger et al., 1996b). This document is a presentation of the results of a number of experiments, realised in the scope of the European program...

Microsatellite markers as a tool to study reproductive success in the Pacific oyster, Crassostrea gigas (Thunberg), under controlled hatchery conditions ArchiMer
Boudry, Pierre; Collet, Bertrand; Cornette, Florence; Hervouet, Veronique; Bonhomme, Francois.
Oysters, like many marine species have a very high fecundity. Previous studies have shown that populations, from both hatcheries and the natural environment, have very low Ne/N ratios. These observations reveal high variation in reproductive success. In order to study individual reproductive success under controlled conditions, we used microsatellite markers to quantify parental contributions in in vitro crosses (5 males and 5 females) of Crassostrea gigas, the Pacific oyster. High polymorphism of the microsatellites (more that 50 alleles per locus) eased the parentage identifications. The results of a cross allowing gametic competition were compared with the results from a second cross where the gametes of the same parents were kept separate for each...

Diversité et différentiation génétique chez l'huître plate européenne : de la population à l'individu (Actes) ArchiMer
Boudry, Pierre; Launey, Sophie; Diaz-almela, A.; Naciri-graven, Yamama; Ledu, Christophe; Mira, S.; Taris, Nicolas; Bonhomme, Francois; Lapegue, Sylvie.
PCR-based DNA markers enable the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European fiat oyster (Ostrea edulis) populations aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species. An Atlantic/Mediterranean differentiation pattern was observed. Similar patterns of variation were found using allozymes, microsatellites and mitochondrial DNA, aIl which support an isolation by distance model. High variability was observed in the range of diversity between populations using a mitochondrial marker, a reflection of small effective population sizes in sorne locations. A 10fold quantitative difference was observed in Fst,...

Variabilite genetique de quelques especes de crevettes Peneides d'interet aquacole ArchiMer
Ko, G; Pasteur, N; Bonhomme, Francois; Liao, D.
Thirteen populations belonging to seven species of Penaeid prawns (Crustacea, Decapoda) were analyzed at 29 protein loci. The authors show that both inter and intrapopulational genetic diversity are very low. These results are discussed in view of their use as guidance for aquaculture programs.
